The alcohol wipe is the "insurance policy" for your manicure. It strips away natural oils and dust that would otherwise sit between your nail and the glue. Without this step, the adhesive attaches to the oil instead of your nail, causing the falsies to pop off prematurely.

Use it as the very last step before applying the glue/sticky tabs. You should have already pushed back your cuticles and filed/buffed your natural nails. Once you wipe a nail, it is officially "prepped", do not touch it with your fingertips, or you will transfer oils back onto the surface.
